چکیده

UNLABELLED PREMISE OF THE STUDY Novel microsatellite markers were characterized in the wind-dispersed and dioecious neotropical tree Triplaris cumingiana (Polygonaceae) for use in understanding the ecological processes and genetic impacts of pollen- and seed-mediated gene flow in tropical forests. • METHODS AND RESULTS Sixty-two microsatellite primer pairs were screened, from which 12 markers showing five or more alleles per locus (range 5-17) were tested on 47 individuals. Observed and expected heterozygosities averaged 0.692 and 0.731, respectively. Polymorphism information content was between 0.417 and 0.874. Linkage disequilibrium was observed in one of the 66 pairwise comparisons between loci. Two loci showed deviation from Hardy-Weinberg equilibrium. An additional 14 markers exhibiting lower polymorphism were characterized on a smaller number of individuals. • CONCLUSIONS These microsatellite markers have high levels of polymorphism and reproducibility and will be useful in studying gene flow and population structure in T. cumingiana.

Characterization of nuclear microsatellite markers for Rumex bucephalophorus (Polygonaceae) using 454 sequencing1

PREMISE OF THE STUDY Nuclear microsatellite markers were developed in Rumex bucephalophorus subsp. canariensis (Polygonaceae) to investigate its genetic diversity and structure. METHODS AND RESULTS Sixteen polymorphic microsatellite markers were obtained using 454 next-generation sequencing with di-, tri-, and tetranucleotide repeats. The average number of alleles was 5.688 and 3.813 for R. b...

Transferability of barley microsatellite markers for assessing genetic relationships of wheat wild relatives of Triticum and Aegilops genera

Comparative genomics has revealed a high degree of collinearity between related species, which allows transferability of interspecies and intergenus   markers. The aim of this study was to examine the transferability of barley microsatellite markers among species of Triticum and Aegilops genus for determination of their genetic relationships. The experiments was carried out in faculty of agricu...

Towards a standard battery of microsatellite markers for the analysis of the Leishmania donovani complex.

The investigation of microsatellite markers has recently superseded that of isoenzymes for many population-biology applications. Microsatellites have the advantages of being dominant, neutral, highly polymorphic and easily scored by high-throughput methods. However, it is necessary to develop a new panel of markers for each group of organisms of interest. Previously, only about 5% of the marker...

Development and characterization of microsatellite markers in the African deciduous tree Terminalia superba (Combretaceae)1

PREMISE OF THE STUDY Microsatellites were designed and characterized in the African timber forest tree Terminalia superba (Combretaceae). Due to their high variability, these markers are suitable to investigate gene flow patterns and the structure of genetic diversity. METHODS AND RESULTS From a genomic library obtained by next-generation sequencing, seven monomorphic and 14 polymorphic micro...

PREMISE OF THE STUDY This study was designed to develop and characterize a preliminary set of nuclear microsatellite markers for Aspidosperma polyneuron, an endangered tropical tree species. METHODS AND RESULTS Leaf samples were collected from a group of mature A. polyneuron trees in a large area along the Pardo and Mogi-Guaçu river basins in São Paulo State in the southeastern region of Braz...
